If you stay only 1 night, best to park at the Palm trolley station then ride the trolley down. Trolley stops at 12:55 PM until 5 AM.

Cascada, Rizo and Ibiza are owned by HK, singles are $85 a night on weekends, slightly cheaper on weekdays.

In La Zona Roja area, hotel Leyva costs $25-35 a night with music thumping all night from Chicago Club, Hacienda de Santiago is 1 small block East, quieter, $35-45, with kitchenette, fridge and free parking. Both are about 1 star. There were reports of roaches and bed bugs but I never found any, but I only stayed 1 night at each.

Hotels Rapido, Cecy, Riviera, Michoacan, Irma ect on Constitucion are all $35, about half star with putas hanging out at the door and in lobby, so sexy but I never stayed over.

I am not sure which crossing has the shorter line, but it's easy enough to use PedEast, which is the traditional crossing and is open 24/7. There are several parking lots near the border. The one behind the Jack in the Box (address appears to be 729 E San Ysidro Blvd) is a big lot with a reasonable rate for overnight parking.

Another option that I found even more convenient is to park at the Palm Ave trolley station (easy, free parking) and take the trolley to San Ysidro station. That will take you right to the border crossing. Just bear in mind the trolley doesn't run from around midnight to 4 am.

Ideally you should have Global Entry or SENTRI so that you don't have to wait in a long line on your way back into the US. Or you can use the bus service that I believe someone mentioned here.

Remember to fill out and print an FMM form in case you get asked for it (you probably won't). When you fill out the form, enter "El Chaparral" as the port of entry (took me forever to figure that out).

The cheap rooms at Cascadas are decent but get a lot of noise from outside. You may want to bring ear plugs if you're a light sleeper. Hotel Rizo across the street is another option (same ownership); I am not sure whether those rooms are quieter.

So Farmacia Roma is on East end of Ave Frontera, across from Ped East taxi stand with a doctor office next to it, even open late, very convenient for the bros seeking quick and convenient health care.
